Basic Information:

1. General Information of Requester

The unit is State Key Laboratory of Trauma, Burns and Combined Injury, Trauma Center, Center of Bone Metabolism and Repair,Institute of Surgery Research, the third hospital affiliated to Army Medical University. There are 16 full-time research and technical personnel in this unit. There is a SPF animal room in the laboratory. This laboratory is mainly engaged in basic and clinical study of bone development, disease (bone metabolic disease, genetic disease) and bone, cartilage damage repair. In addition to cell culture room and conventional molecular biology and histopathology, the lab has the technical platform of transgenic mice which was built for ES cell and DNA microinjection.

 

2. International Cooperation Status

In cooperation with Dr. Chuxia Deng from NIH in the United States to study the roles of FGFRs in skeletal development and diseases; In cooperation with Dr. Jerry Feng in Texas A&M Baylor College of Dentistry Baylor to study the roles of FGF23DMP in the bone development In cooperation with Dr. Chuanju Liu in New York University to carry out the roles of p204FGFR3 in the cartilage development and degenerative diseases In cooperation with Dr. Di Chen to carry out the study of the regulation of FGFR3 in bone formation.

1. Importance and necessity of requested project, and relations with key projects and industries of Chongqing .

In modern society, aging (osteoporosis), trauma and other reasons lead to the high incidence of fracture. Although some new therapies, such as internal fixation, have increased the level of clinical treatment, not a few patients also have bone defects, delayed healing, and even bone nonunion. These complications seriously affect the patient's self-care ability and social life, bringing great burden to the family and society. Therefore, it is an important problem to study the role and mechanism of bone and joint injury and degeneration, and to promote the repair of bone and joint injury.

 

2. Technology breakthroughs and innovations of requested project

Establishment of genetically engineered mouse models, and the preparation of bone and joint injury and degeneration models; and to study the roles of Sirt proteins in bone and joint injury and degeneration; to observe whether interventions with the regulation of Sirt proteins using RNAi and Crispr/Cas9 can promote bone and joint injury repair and relieve bone and joint degeneration.
